Abstract
The effects of LSD (Delysid, Sandoz) on chromosome damage and on the ability of mammalian cells to repair UV light-induced damage were investigated. Methods Concanavalin-A-stimulated human peripheral blood lymphocytes or human fibroblasts were incubated with LSD 0.5-2.5 mcg/ml and 10-3 8.3mcg/ml respectively. Metaphase -rich cultures were harvested for chromosomes and each case analyzed for gap, breakage polyploidy, fragmentation, endoreduplication and total abnormalities Peripheral blood was obtained from 7 LSD users or 17 controls and lymphocyte suspensions exposed to UV light. The cells were then incubated with 3H-thymidine and autoradiographic analyses performed. DNA repair was measured as the 0f cells showing repair (25 grains labeling or less). Normal human lymphocytes were incubated with LSD, ethidium bromide(which intercalates in DNA) and caffeine, then exposed to UV light and DNA repair assessed after incubation with tritiated 3H-thymidine as before. LSD had no significant effects on human lymphocyte chromosomes or on human skin fibroblast chromosomes in vitro. LSD in vivo and in vitro did not impair the ability of DNA to repair UV damage. Caffeine induced a 50% reduction in repair labeling. Ethidium bromide induced a 29.3%reduction in repair labeling but this appeared to be due to its effect on enzymes and not to inter - calation in the DNA
