Abstract
The effects of chlorimipramine (C) and LSD on efflux of precursor-formed 3H-serotonin were studied. Methods - Male albino Charles River rats (220-280 g) were anesthetized with chloral hydrate and placed in a stereotaxic apparatus. A cerebroventricular perfusing technique was used to determine the efflux of 3H-5-HT synthesized in vivo from intraventricular 3H-tryptophan (Schwarz-Mann, 10 mcC/mcl). Impulse flow in sero-toninergic neurones was measured by recording single unit activity from raphe (5-HT) neurones. The effects of C (Anafranil, CIBA-Geigy) and d-LSI) given i.p. were determined. C 5 mg/kg completely inhibited raphe cell firing for about 1 hr. as did LSD 75 mcg/kg. This inhibition was not associated with a reduction in action potential size. LSD significantly reduced the efflux of 3H-5-HT (P < 0,025) at a dose of 75 mcg/kg. Increasing the doee to 300 1lg/kg did not have a further inhibitory effect. During the perfusion period, the efflux remained below the control level. C 5 mg/kg did not decrease 5-HT efflux, but at 20 mg/kg there was a significant increase in efflux (P< 0.05), which returned to baseline values after about 90 min. Ro 4-4602 (Roche) 800 mg/kg i.p. děd not significantly effect the rate of 3H-5-HT efflux, indicating that 5-HT synthesis was neglible.
