Abstract
The effects of chronic administration of LSD on serotonin turnover were studied in various regions of the rat brain in vivo. Male rats were treated with i.p.LSD at 20 mcg/kg/day for 28-31 days or with saline. 24 hr after the last dose, brains were removed and separated into pons/medulla,' midbrain, cortex, and forebrain; the diencephalon and striatum were treated as a single region. The serotonin:(5HT) and 5-hydroxyindoteacetic acid (5HIAA)' concentrations and the rates of turnover of these compounds were-'studied as described previously following pulse labeling doses of 3H-tryptophan. The effects of acute doses of 500 mcg/kg i.p. Of LSD given 3 and 1 hr before death on brain 5HT and 5HIAA were also determined in a similar manner. Studies of 5HT conversion to tyramine were also performed. The 5HT levels in pons/medulla, midbrain, forebrain and cortex were not affected after 28-31 days treatment wish LSD. The level of 5HIAA in the pons/medulla- was slightly increased and that -in the midbrain was significantly reduced. 5HIAA levels in forebrain and cortex were not altered by chronic LSD treatment. After acute treatment with LSD the 5HT levels in brainstem, forebrain and cortex were all increased by about 50%, suggesting a reduced turnover in these regions. After chronic LSD treatment and pulse labeling the levels of 3H-5HT in pons/medulla, midbrain and cortex but not forebrain, were increased. Separation of tyramine from 5HT showed that less than 50f the radioactivity was converted to tyramine in this study. Chronic administration of LSD has only small effects on the turnover of serotonin in the rat brain in vivo.
