Abstract
The effects of LSD on in viva leucine (Len) incorporation into different areas of the rat brain were studied. Methods Adult male Wistar rats were given (3H)-Leu (8.33 mCi, i.p.) with or without pretreatment with LSD (1 ng/kg, i.m.) 1 hr previously. Rats were killed, under ether narcosis 1 or 7 hr after receiving (3H)-Leu and the left ventricle was perfused with 100 ml Ringer solution (at body temperature) containing Liquemin and 6 0lutaraldehyde. After embedding in paraffin sections of brain were prepared and examined by autoradiography. The numbers of silver grains were counted in 6 layers of the parietal cortex, 4 layers of Ammon's horn, the corpus callosum and the cortex, medulla end lateral nucleus of the cerebellum. Results In brain slices prepared 1 hr after injection of (3H)-Leu, LSD reduced incorporation of isotope into all layers of the parietal cortex, particularly those with high densities of nerve cells and glial cells. This effect of LSD was somewhat reduced 7 hr after giving (3H)-Leu. In contrast, LSD pretreatment increased the incorporation of (3H)-Leu into all layers of the cerebellum 1 hr after label. In Ammon's horn and the dentate gyrus, LSD treatment also increased incorporation of (3H)-Leu and this effect was greater in rats killed 7 hr. rather than 1 hr. after injection of (3H)-Leu. The effects of LSD on brain metabolism, and the ability of chlorpromazine to prevent these effects, are discussed together with the effects of other drugs such as imipramine, desmethylimipramine and mescaline.
