Abstract
Single dose (i.v.) effects of MES (4 mg/kg), PSI (0.05 mg/ kg) and DMT (0.3 mg/kg) were evaluated in nontolerant and LSD tolerant chronic spinal dogs. Appetite suppressant effects of MES and PSI were assessed In intact dogs. In nontolerant dogs, MES, PSI and DMT specifically resembled LSD (15 mcg/kg) and not d-amphetamine (A) (2 mg/kg) with respect to their physiologic and behavioral effects. LSD tolerant dogs (30 v9 LSD/kg/day) were cross tolerant to PSI and MES but not A. Partial cross tolerance to DMT developed as its mydriasis and facilitative effect on the flexor reflex were not significantly reduced. These results are in agreement with clinical pharmacologic studies. LSD, MES and PSI administered s.c. suppressed food intake in dog and produced parallel dose-response curves. However, their slopes were significantly less than that of A, LSD, MES and PSI did not completely abolish food intake as did A. In conclusion MES, PSI and DMT are pharmacologically equivalent to LSD in the dog as they are in man suggesting that the dog can be used to assess compounds for LSD-like activity.
