Abstract
The partial agonist activity of presumptive 5-HT antagonists was examined in the rat. Method Rats were trained for 3 mth to discriminate LSD (0.16 mg/kg, i.p.) from saline and to respond by pressing 1/2 bars for a food reward. Test performance was monitored 60 min after administra tion of cyproheptadine (1.25. 2.5, 5 and 10 mg/kg. s.c.), methy sergide (0.16. 0,63, 2.5 and 10 mg/kg, s.c.) and mianserin t2.5 10 and 40 mg/kg, s.c.). The agonist specificity of the LSD cue was confirmed by i.p. injection of mescaline (5-40 mg/kg) or quipazine (1.25-5 mg/kg) 15 minbefore the test. Results Mescaline and quipazine produced a dose-dependent increase in the number of rats selecting the LSD lever. Cyproheptadine and methysergide produced partial generalization with LSD. By contrast, mianserin produced a monophasic dose-response curve - and a higher maximum effect. Cyproheptadine, methysergide and mianserin may have partial agonist effects at 5-HT receptor sites in the rat CNS.
