Abstract
The effect of indoleamine or phenylethylamine hallucinogens on serotonin levels in cell bodies of dorsal and median raphe nuclei was studied in rats. Method Male Sprague-Dawley rats (140-160 g) received saline, 100 mcg/kg d-LSD, 2.5 mg/kg N,N-dimethyltryptamine, 1 mg/kg 25-dimethoxy-4-methylamphetamine or 20 mg/kg mescaline i.p. Rats were sacrificed and 3 brain regions prepared:, from the mid-brain raphe nuclei: dorsal B7, a cluster of cells below the aqueduct of Sylvius; ventromedial B7, and B8 - a scattered group of cells below the decussation of the superior cerebellar peduncle. From each region, 10-15 intra- and extra- cellular readings were taken. Re suits ANOVA analysis revealed 3 significant effects of drug treatments on intracellular fluorescence fading measure in all 3 regions. Dorsal B7 F= 4.79, df = 4,18. Ventral B7, F=4.25 df = 4,18 and B8, F=5.61, df = 4,18. The only region in which a significant effect was found with extracellular measurement was dorsal B7. Each of the test drugs increased intracellular 5-HT in all 3 regions. Both indoleamine and phenylethylamine hallucinogens are similar in their effects on major mid brain raphe nuclei.
