Abstract
The mechanisms of action of three classes of psychoactive drugs— entactogens, hallucinogens, and anorectics—were investigated at the molecular, cellular, and behavioral levels. The selectivity of analogs of the entactogen 3,4-methylenedioxy-amphetamine in interacting with monoamine neurotransmitter carrier mechanisms is governed by the position and orientation of the phenyl ring etherlinked oxygen unshared electrons. The catecholamine mechanisms display more rigorous structural requirements than the serotonin mechanism, and this likely reflects a requirement for hydrogen bonding interactions with the catecholamine carrier proteins.
